Adenosine Deaminase Severe Combined Immunodeficiency (ADA-SCID) is a rare yet life-threatening genetic disorder. Our recent study evaluates the prevalence, diagnostic advancements, and evolving therapies for ADA-SCID, offering strategic insights to improve management and patient outcomes.

July FDA Approval Snapshot

Sephience™ (sepiapterin) Oral Powder received FDA approval on July 28, 2025 for treating phenylketonuria (PKU) in both children and adults aged 1 month and older.

⚙️ Mechanism of Action:

Sepiapterin works via a dual mechanism Synapse+6

▪️A precursor to tetrahydrobiopterin (BH₄), the essential cofactor for phenylalanine hydroxylase (PAH), increasing BH₄ intracellularly.

▪️Acts as a pharmacological chaperone, stabilizing misfolded PAH variants and enhancing enzymatic activity even in BH₄‑insensitive mutations.

By restoring PAH function, it effectively lowers blood phenylalanine (Phe) levels.

✅ Indications

Hyperphenylalaninemia (HPA) in adult and pediatric patients (≥ 1 month) with sepiapterin‑responsive phenylketonuria (PKU)

⚠️ Contraindications

Known hypersensitivity to sepiapterin or any excipient in the formulation (standard for metabolic therapies; specific labeling not publicly detailed yet).

⚠️ Common Side Effects (from clinical trials and PI)

▪️Upper respiratory tract infection
▪️Diarrhea
▪️Headache
▪️Abdominal pain
▪️Hyperphenylalaninemia (low phenylalanine levels)
