Difference between Antibiogram Test with Salt Water and the standardized laboratory method

An antibiogram test is used to determine the sensitivity of bacteria to various antibiotics. It is typically conducted using standardized laboratory methods, such as the disc diffusion (Kirby-Bauer) method, on nutrient-rich agar media like Mueller-Hinton agar.

In this test, bacterial suspensions are usually prepared in sterile normal saline (0.85–0.9% NaCl) to match a standardized turbidity (0.5 McFarland), ensuring uniform growth.

Using salt water (e.g., seawater or high-salt solutions) instead of standard saline or media is not recommended, as:

High salinity may inhibit bacterial growth or alter antibiotic activity.

It lacks essential nutrients for bacterial replication.

It can lead to inaccurate results in antibiotic susceptibility testing.

Conclusion: While sterile saline is used to prepare bacterial suspensions, salt water (especially seawater or hypertonic solutions) is not suitable for performing an accurate antibiogram test.
